Micro/nanofluidics has emerged in recent years as a powerful method of manipulating fluids at small length-scales. To highlight their versatility and great potentials, I present 2 simple nano/microfluidic platforms to showcase how nano- and microfabrication techniques can be coupled with fluidics for nanoplasmonic biosensing and immunoassay applications.

(a) Nanoplasmonic platforms for multiple biosensing applications: We develop a nanoplasmonic platform with remarkable biocompatibility, for long-term survival of prokaryotic and eukaryotic cells, thereby enabling nanoplasmonic based cell bioassays. This nanoplasmonic platform is used for monitoring mitosis of fibroblasts for 7 days, E. coli biofilm formation, and DNA polymerase activity in real-time. In addition, the same nanoplasmonic platform is used to conduct routine protein/DNA based immunoassays with a limit of detection of∼66 zM.

(b) Making Waves in a Total Capture Nanofluidic Immunoassay: understanding nanoconfinement phenomena is necessary to develop nanofluidic technology platforms. We demonstrate a convection limited nanofluidic immunoassay that achieves total capture of a target analyte, and an apparent shift in the reaction equilibrium due to nanoconfinement.

Amy Shen leads the Micro/Bio/Nanofluidics Unit at Okinawa Institute of Science and Technology in Japan. She was a tenured faculty member in Mechanical Engineering at University of Washington before moving to Japan in 2014. Her research is focused on nano/microfluidics with applications in biotechnology. Amy is a Member-at-Large of the Society of Rheology Executive Committee (2018-2020), a delegate of the individual Members to the European Society of Rheology Committee (2019-2023), an honor member of Phi Kappa Phi and Pi Tau Sigma. She received the Ralph E. Powe Junior Faculty Enhancement Award in 2003 and the National Science Foundation's CAREER Award in 2007, and she held a Fulbright Professorship in 2013. Amy gave the 2019 Bergveld lecture at the University of Twente. Amy is an associate editor for Micomachines, Biomedical Microdevices, and Scientific Reports, and on the editorial advisory board of VIEW.
